The reason that a USB/ Flash drive can not be partitioned in Windows is because of the fact that these drives are shown as removable media (external drives). So for usb drive partition, we have to first show it as internal or fixed media to Windows and then we can partition and use it as a regular hard drive.
Lexar’s USB format tool is best tool out there that is designed to work for Lexar usb drives but the good thing is that it works with almost all brands USBs. The tool is portable so it is easy to carry and no need to install it. The tool not only works flipping the removable media thing from USB but is also able to format usb/flash drives, setting the label of usb and partition them. You only need to press the flip removable bit button if you want to prepare the flash drive for use with multiple partitions. You also need to remove and reconnect usb drive to your computer.
It should be noted that Lexar USB Format will not partition the usb flash drive which has to be done with other tools, you can use the built in partition tool in the operating system for that ( Partitioning in Vista will help you if you are running Windows Vista.)
